A interleucina 8 (IL-8 ou IL8), tamén chamada CXCL8, é unha quimiocina producida polos macrófagos e outros tipos celulares como as células epiteliais, células do músculo liso das vías aéreas[1] e células endoteliai. As células endoteliais almacenan IL-8 nas súas vesículas de almacenamento chamadas corpos de Weibel-Palade.[2][3] Nos humanos, a proteína interleucina 8 está codificada no xene IL8 do cromosoma 4.[4] A IL-8 é producida inicialmente como un péptido precursor de 99 aminoácidos, que posteriormente sofre unha clivaxe para dar lugar a varias isoformas activas de IL-8.[5] En cultivo, a principal forma segregada polos macrófagos é un péptido de 72 aminoácidos.[5]

Hai moitos receptores na superficie da membrana plasmática aos que se pode unir a IL-8; os tipos que se estudan máis frecuentemente son os receptores serpentina acoplados á proteína G CXCR1 e CXCR2. A expresión e afinidade pola IL-8 difire entre estes dous receptores (a de CXCR1 é maior ca a de CXCR2). Función

A IL-8 ten dúas funcións. Induce a quimiotaxe en células diana, principalmente en neutrófilos e outros granulocitos (por iso tamén se lle chama factor quimiotáctico de neutrófilos), causando que estes migren cara ao sitio de infección. A IL-8 induce tamén a fagocitose unha vez que estas células chegaron ao seu destino. A IL-8 tamén é un potente promotor da anxioxénese. Nas células diana, a IL-8 induce unha serie de respostas fisiolóxicas necesarias para a migración e a fagocitose, como un incremento do Ca2 + intracelular, a exocitose (por exemplo, a liberación de histamina), e a explosión respiratoria.

A IL-8 pode ser segregada por calquera célula que teña receptyores de tipo Toll, que están implicados na resposta innata inmunitaria. Usualmente, os macrófagos son os que detectan primeiro un antíxeno, polo que son as primeiras células que liberan IL-8 para recrutar outras células. Tanto as formas monómeras coma as homodímeras da IL-8 son potentes indutores dos receptores de quimocinas CXCR1 e CXCR2. O homodímero é máis potente, pero a metilación da Leu 25 pode bloquear a actividade dos homodímeros.

Crese que a IL-8 desempeña unha función na patoxénese da bronquiolite, que é unha doenza común do tracto respiratorio causada por infeccións virais.

Segundo algúns estudos, se unha muller preñada ten niveis altos de interleucina 8, aumenta o risco de esquizofrenia na súa descendencia.[13] Ter niveis altos de IL-8 reduce a probabilidade de ter respostas positivas á medicación antipsicótica na esquizofrenia.[14]

Nomenclatura

A IL-8 foi renomeada como CXCL8 polo Subcomité de Nomenclatura de Quimiocinas da Unión Internacional de Sociedades Inmunolóxicas,.[15] O seu símbolo xénico aprobado pola Organización do Xenoma Humano (HUGO) é CXCL8.

Regulation of Expression

The expression of IL-8 is negatively regulated by a number of mechanisms. MiRNA-146a/b-5p indirectly represses IL-8 expression by silencing the expression of IRAK1.[16] Additionally, the 3'UTR of IL-8 contains a A/U-rich element that makes it extremely unstable under certain conditions. IL-8 expression is also regulated by the transcription factor NF-κB.[17] NF-κB regulation represents a novel anti-IL-8 therapy for use in inflammatory diseases such as cystic fibrosis.
